Results

Thirty-two patients with schizophrenia were randomized to receive placebo, low dose pregnenolone (100 mg), rapid escalation to high dose pregnenolone (500 mg) and slow escalation to high dose pregnenolone (500 mg) given adjunctively to antipsychotics. The group receiving rapid escalation to high dose pregnenolone showed significant improvements in PANSS total scores, PANSS negative scores, SANS total scores. Pregnenolone also seemed to improve verbal memory and sustained attention. Pregnenolone is a precursor of steroid hormones and is a glutamate NMDA receptor agonist.
